本文报道一种同时快速测定合成肽中色氨酸和酪氨酸含量的二阶导数光谱法,本法不需要水解或衍生化反应程序。试剂光谱分析用的L-色氨酸和L-酪氨酸为分析纯。紫外光谱分析采用Hewlett-Packard8452A型分光光度计二阶导数光谱法记录吸收度值。将合成肽溶解在0.1NNaOH溶液中,制成约200nmol/ml浓度的溶液,分别以0.1NNaOH配制相同浓度的色氨酸和酪氨酸标准溶液为对照进行扫描。
In this paper, we report a second-order derivative spectroscopy method for rapid and simultaneous determination of tryptophan and tyrosine in synthetic peptides. This method does not require hydrolysis or derivatization reactions. L-Tryptophan and L-Tyrosine were used as analytically pure reagents for spectral analysis. UV spectral analysis Absorbance values were recorded by second order derivative spectroscopy using a Hewlett-Packard model 8452A spectrophotometer. The synthesized peptide was dissolved in 0.1N NaOH solution to make a solution with a concentration of about 200 nmol / ml, and the same concentration of tryptophan and tyrosine standard solution was prepared as a control by using 0.1N NaOH respectively.
