目的:建立同时测定维吾尔药材天山花楸中金丝桃苷和芦丁含量的RP-HPLC分析方法。方法:采用HPLC法,色谱柱为Symmetry shield RP8(5μm,4.6 mm×150 mm);流动相为甲醇-0.05%磷酸(三乙胺调pH至3.0)(40∶60),柱温40℃,流速1.3 mL·min-1,检测波长360 nm。结果:金丝桃苷和芦丁分别在20.0~100.0 ng和15.8~79.0 ng范围内,进样量与色谱峰积分面积呈良好线性关系,相关系数分别为0.9997和0.9999,平均回收率(n=6)分别为102.3%和100.5%。结论:应用本法测定金丝桃苷和芦丁的含量稳定可靠,重复性好,可用于天山花揪药材的质量控制。
OBJECTIVE: To establish an RP-HPLC method for the simultaneous determination of hyperin and rutin in Uighur medicinal herbs. Methods: HPLC method was used. The chromatographic column was Symmetry shield RP8 (5μm, 4.6 mm×150 mm). The mobile phase consisted of methanol-0.05% phosphoric acid (triethylamine adjusted to pH 3.0) (40:60). The column temperature was 40°C. The flow rate was 1.3 mL·min-1 and the detection wavelength was 360 nm. RESULTS: Hyperin and rutin were in the range of 20.0-100.0 ng and 15.8-79.0 ng, respectively. The injection volume was in a good linear relationship with the integral area of the chromatographic peak. The correlation coefficients were 0.9997 and 0.9999, respectively. The average recovery rate (n= 6) 102.3% and 100.5% respectively. Conclusion: The application of this method to determine the content of hyperin and rutin is stable and reliable, with good repeatability. It can be used for the quality control of the medicinal materials of Tianshan Huayu.
