目的研究不同时间不同剂量白细胞介素-4(IL-4)作用下A549细胞钠离子通道β亚基(β-ENaC)的变化情况。方法以A549细胞为模型,在含IL-4培养基不同时间不同剂量培养下,分别用免疫印迹法以及RT-PCR法测定β-ENaC蛋白表达以及mRNA变化转录情况。结果IL-4可以呈剂量、时间依赖性的下调β-ENaC的表达,这种蛋白表达的下调作用是通过基因调控起作用的。IL-4(1 ng/ml)培养A549细胞3 h,β-ENaC蛋白及mRNA开始下降,在6 h后出现显著差异(P<0.05),不同剂量的IL-4(0.1、10、100 ng/ml)培养A549细胞12 h在0.1 ng/ml时即出现显著的差异(P<0.05)。结论IL-4可通过下调β-ENaC的表达抑制肺泡水肿液清除能力,从而不利于急性肺损伤/急性呼吸窘迫综合征的预后。
Objective To investigate the changes of sodium ion channel β subunit (β-ENaC) in A549 cells under different doses of interleukin-4 (IL-4). Methods A549 cells were used as a model to detect the expression of β-ENaC and the transcriptional changes of mRNA in different doses of IL-4 culture medium by Western blotting and RT-PCR respectively. Results IL-4 down-regulated the expression of β-ENaC in a dose-dependent and time-dependent manner. The down-regulation of IL-4 expression was mediated through gene regulation. A549 cells were cultured with IL-4 (1 ng / ml) for 3 h, the protein and mRNA expression of β-ENaC began to decline, and the difference was significant after 6 h (P <0.05) / ml) cultured A549 cells 12 h at 0.1 ng / ml significant difference (P <0.05). Conclusion IL-4 can inhibit the clearance ability of alveolar edema by down-regulating the expression of β-ENaC, which is not conducive to the prognosis of acute lung injury / acute respiratory distress syndrome.
