作者报道伤寒杆菌温度敏感株(ts)51-1作为口服活菌苗的Ⅰ期研究结果。 菌苗为ts变异株51-1的液体制剂,接种对象为3名健康男青年。受试者每次口服含2~3×10~9个ts51-1变异株活菌的菌苗。隔日1次,连续3次。每名受试者先服1.3%NaHCO_3溶液120ml以中和胃酸,约l~5分钟后,再口服30ml1.3%NaHCO_3溶液加1ml活菌苗的混合液。 用ELISA法测定抗伤寒杆菌O、H和Vi抗原的血清抗体。加热灭活的野型Ty2伤寒杆菌作为O抗原来源。野型Ty2伤寒杆菌的福尔马林固定的能动株作为H抗原来源,活ts51-1株作为Vi抗原来源。用分泌性IgA(sIgA)特异性抗血清测定sIgA抗体。
The authors report the temperature-sensitive strain of typhoid fever (ts) 51-1 oral live vaccine as a phase Ⅰ study. The bacterin was a liquid preparation of ts mutant strain 51-1, which was administered to 3 healthy young men. Subjects were orally treated with 2 ~ 3 × 10 ~ 9 strains of ts51-1 mutant live bacteria vaccine. 1 times every other day for 3 consecutive times. Each subject first served 120ml of 1.3% NaHCO3 solution to neutralize gastric acid, about l ~ 5 minutes later, and then oral 30ml1.3% NaHCO3 solution plus 1ml of live vaccine mixture. Serum antibodies against Salmonella typhimurium O, H and Vi antigens were determined by ELISA. Heat-inactivated tyrosinase-negative Ty2 bacterium was used as a source of O antigen. Wild-type Ty2 typhoid bacilli formalin-fixed active strain as a source of H antigen, live ts51-1 strain as Vi antigen source. SIgA antibodies were measured using secreted IgA (sIgA) specific antisera.
