目的:在改良的EuroColins心脏保存液(mEC液)中添加黄芪皂甙(AS),观察黄芪皂甙对心脏保存的作用。方法:将Wistar大鼠分成mEC组和AS组。分别用mEC液和含AS的mEC液简单冷保存离体大鼠心脏6h后,用Langendorf离体鼠心模型再灌注30min,研究保存后的心功能及抗氧自由基损伤作用。结果:AS组与mEC组比较,保存后心脏的心功能和冠脉流量明显好于mEC组(P<005),心肌含水量显著降低(P<001),心肌细胞乳酸脱氢酶和肌酸激酶的漏出量显著减少(P<005和P<001),心肌细胞超氧化物歧化酶活性显著增高(P<001),而过氧化脂质含量明显降低(P<005)。结论:黄芪皂甙能提高mEC液的心脏保存效果,可能与负性肌力和抗氧自由基损伤作用有关
OBJECTIVE: To observe the effect of astragaloside on the preservation of the heart by adding astragaloside (AS) to the improved EuroColins cardiac preservation solution (mEC fluid). Methods: Wistar rats were divided into the mEC group and the AS group. The isolated rat hearts were cryopreserved in mEC fluid and ASEC-containing myc fluid for 6 hours and then reperfusioned for 30 min using a Langendorf isolated rat heart model to study the preserved heart function and anti-oxidative free radical damage. RESULTS: Compared with the mEC group, the cardiac function and coronary flow in the AS group were significantly better than those in the mec group (P < 0.05), and the myocardial water content was significantly lower (P < 0.01). The leakage of enzyme and creatine kinase was significantly reduced (P<005 and P<001), the activity of superoxide dismutase in myocardial cells was significantly increased (P<001), and the lipid peroxide content was significantly reduced. (P<005). Conclusion: Astragalus saponins can improve the cardiac preservation effect of mEC fluid, which may be related to the negative inotropic and antioxidant free radical damage
