目的探讨油烟中的细颗粒物(PM2.5)对原代培养的胎鼠肺泡Ⅱ型上皮细胞(AECⅡ)DNA的损伤效应。方法将1只妊娠18 d的SPF级ICR小鼠体内的胎鼠肺组织制成AECⅡ细胞悬液,取对数生长期细胞,调整细胞密度为1×106/ml,分别加入终浓度为0(溶剂对照,含10%FBS的DMEM)、12.5、25、50、100μg/ml的PM2.5(来源于烹调油烟)溶液,培养6、12 h后进行MTT试验和彗星试验,并检测AECⅡ细胞的尾长、尾部DNA百分比、尾矩和Olive尾矩。结果与溶剂对照组比较,50、100μg/ml PM2.5染毒6、12 h时胎鼠AECⅡ存活率下降,差异均有统计学意义(P<0.05);12.5、25、50μg/ml PM2.5染毒6、12 h时胎鼠AECⅡ的尾长、尾部DNA百分比、尾矩和Olive尾矩升高,差异均有统计学意义(P<0.05)。随着PM2.5染毒浓度的升高,AECⅡ细胞存活率呈下降趋势,尾长、尾部DNA百分比、尾矩和Olive尾矩均呈上升趋势。结论油烟中的PM2.5可降低AECⅡ的细胞活性,对AECⅡ的DNA具有损伤效应。
Objective To investigate the effect of particulate matter (PM2.5) in soot on the DNA damage of fetal rat alveolar type Ⅱ epithelial cells (AECⅡ). Methods Fetal lung tissue of SPF grade ICR mice on day 18 of gestation was made into AEC Ⅱ cell suspension. The cells in logarithmic growth phase were taken and the cell density was adjusted to 1 × 10 6 cells / ml. The final concentration was 0 ( Solvent control, DMEM containing 10% FBS), 12.5, 25, 50, 100μg / ml PM2.5 (derived from cooking fume) solution. After cultured for 6 and 12 h, MTT assay and comet assay were performed. Tail length, tail DNA percentage, tail moment and Olive tail moment. Results Compared with the solvent control group, the survival rate of AECⅡ in fetal rat decreased at 50,100μg / ml PM2.5 exposure for 6 and 12 h (P <0.05), and 12.5,25 and 50 μg / ml PM2. Tail length, percentage of tail DNA, tail moment and Olive tail moment of fetal rat AECⅡwere significantly increased at 6,12 h (P <0.05). With the increase of PM2.5 concentration, the survival rate of AECⅡ cells decreased. The length of tail, tail DNA, tail moment and Olive tail moment all showed an increasing trend. Conclusion PM2.5 in soot can reduce the cell activity of AECⅡand have damaging effects on DNA of AECⅡ.
