目的 研究鬼臼酰肼哌啶氮氧自由基腙 (GP1)和阿霉素 (DOX)协同抗肿瘤作用。方法 用MTT比色法观察GP1、DOX对K56 2细胞生长的抑制作用。结果 GP1和DOX单独应用时 ,最大浓度 (10 μg·ml- 1)对K56 2细胞的抑制率分别为 2 0 5%和 4 6 0 % ,当用GP11μg·ml- 1和不同浓度的DOX联合应用时 ,对K56 2细胞的抑制作用明显增强 ,DOX的IC50 为 3 87μg·ml- 1,是单用DOX(IC50 为 14 32 μg·ml- 1)的 1/3 7。联合用药的抑制率大于单独用药的总和 ,q >1 15。结论 GP1和DOX两药合用有协同抗肿瘤作用。
Objective To study the synergistic antitumor effect of podophyllohydrazide (GP1) and doxorubicin (DOX). Methods The inhibitory effect of GP1 and DOX on the growth of K562 cells was observed by MTT assay. Results When GP1 and DOX were used alone, the inhibition rate of K56 2 cells at the maximum concentration (10 μg·ml - 1) was 2 0 5% and 46%, respectively, when combined with GP 11 μg·ml - 1 and different concentrations of DOX. When applied, the inhibitory effect on K562 cells was significantly enhanced. The IC50 of DOX was 387 μg·ml-1, which was 1/3 of that of DOX alone (IC50 of 1432 μg·ml-1). The inhibition rate of the combination was greater than the sum of the individual drugs, q >1 15. Conclusion The combination of GP1 and DOX has a synergistic anti-tumor effect.
