目的 :建立人血清中异环磷酰胺的 HPL C测定法。方法 :取血清 1.0 ml,以环磷酰胺为内标 ,用氯仿提取 ,在 6 0℃水浴条件下氮气挥干 ,残渣用 10 0 μl流动相复溶 ,进样 2 5 μl。在 Hypersil- C4 色谱柱 (4.6 mm× 2 5 0 mm,5μm) ,以乙腈 -水 (35∶ 6 5 ,v/ v)为流动相 ,流速为 1.0 m l/ min;检测波长 2 0 0 nm。结果 :本方法在 10~ 2 0 0μg/ ml范围内线性良好 ,相关系数 r =0 .9994 (n =5 )。检测限为 5 μg/ ml。高、中、低三种浓度的绝对和相对回收率分别为84 .32 %~ 88.14 % ,95 .0 0 %~ 98.10 % ,日内和日间 RSD分别为 0 .74 %~ 2 .2 1% ,1.6 0 %~ 2 .4 0 %。结论 :本方法灵敏、特异、简便 ,可用于临床异环磷酰胺血药浓度检测
OBJECTIVE: To establish an HPL C assay for ifosfamide in human serum. Methods: Take 1.0 ml of serum and take cyclophosphamide as internal standard. Extract with chloroform and dry it under nitrogen at 60 ℃. Re-dissolve the residue with 100 μl of mobile phase and inject 25 μl into the sample. On a Hypersil-C4 column (4.6 mm × 250 mm, 5 μm), acetonitrile-water (35:65, v / v) was used as the mobile phase at a flow rate of 1.0 mL / min and the detection wavelength was 200 nm. Results: The method was linear in the range of 10 ~ 200 μg / ml with a correlation coefficient of 0.9994 (n = 5). The detection limit is 5 μg / ml. The absolute and relative recoveries of high, middle and low concentrations were 84.32% -88.14% and 95.0% -98.10%, respectively. The intra-day and inter-day RSD were 0.74% -2.21% , 1.6 0% ~ 2.04%. Conclusion: The method is sensitive, specific and simple and can be used for the determination of ifosfamide serum concentration
