目的探讨Rock-1的表达对宫颈癌侵袭能力的影响。方法采用RT-PCR和Westernblot方法检测正常宫颈组织、宫颈癌细胞株Hela及其以Rho激酶特异性抑制剂Y-27632处理后的Rock-1和磷酸化cofilin表达变化。并用Transwell小室体外侵袭方法,检测Y-27632处理前、后Hela细胞体外侵袭能力的变化。结果宫颈癌细胞株Hela细胞中,Rock-1和磷酸化cofilin的表达量均高于正常宫颈组织(P<0.05)。不同浓度的Y-27632阻断Rock-1作用后,Hela细胞中磷酸化cofilin的表达量及体外侵袭能力降低(P<0.05)。结论 Rock-1在宫颈癌细胞中的表达高于正常宫颈组织;抑制Rock-1的表达能降低宫颈癌细胞株Hela细胞的体外侵袭能力。
Objective To investigate the effect of Rock-1 expression on the invasiveness of cervical cancer. Methods The expression of Rock-1 and phosphorylated cofilin in normal cervical tissue, cervical cancer cell line Hela and Y-27632, a Rho-kinase specific inhibitor, were detected by RT-PCR and Western blot. Transwell chamber invasion assay was used to detect the in vitro invasiveness of Hela cells treated with Y-27632. Results The expression of Rock-1 and phosphorylated cofilin in cervical cancer cell line Hela was higher than that in normal cervical tissues (P <0.05). Different concentration of Y-27632 blocked the effect of Rock-1, the expression of phosphorylated cofilin in Hela cells and in vitro invasive ability decreased (P <0.05). Conclusion The expression of Rock-1 in cervical cancer cells is higher than that in normal cervical tissues. Inhibition of Rock-1 expression can reduce the in vitro invasiveness of cervical cancer cell line Hela.
