目的:观察丝瓜提取物对衰老小鼠学习记忆能力、脑组织形态学及免疫功能的影响。方法:经筛选的昆明种小鼠随机分为对照组、衰老组、丝瓜提取物187.5 mg/kg、375 mg/kg、750 mg/kg 3个剂量组。除对照组以外,均在颈背部皮下持续注射D-半乳糖复制衰老模型,利用跳台实验及Morris水迷宫测试各组小鼠学习记忆能力,脏体比方法测定各组小鼠脑指数、胸腺指数和脾脏指数,ELISA法检测各组小鼠血清中细胞因子IL-2,IFN-r的浓度和T淋巴细胞增殖活性,HE染色法观察各组小鼠脑组织形态学变化。结果:衰老组在跳台实验的能力测试中错误次数增多;在水迷宫测试中逃避潜伏期延长,平台象限游泳时间缩短,穿越平台次数减少;脑组织细胞数量明显减少;脑指数、胸腺指数和脾脏指数减少;IL-2、IFN-r水平和T淋巴细胞增殖活性降低;与衰老组比较,丝瓜提取物(375 mg/kg、750mg/kg)可明显拮抗上述指标变化。结论:丝瓜提取物可以改善衰老小鼠的学习记忆,同时增加机体免疫活性,达到延缓衰老进程的作用。
Objective: To observe the effect of loofah extract on learning and memory ability, brain morphological and immune function in senile mice. Methods: Kunming mice were randomly divided into three groups: control group, aging group, loofah extract 187.5 mg / kg, 375 mg / kg and 750 mg / kg. D-galactose replication senescence model was injected subcutaneously in the back of the neck except for the control group. The learning and memory abilities of the mice in each group were tested by jumping test and Morris water maze test, and the brain index, thymus index And spleen index. Serum levels of IL-2, IFN-γ and T lymphocyte proliferation were detected by ELISA. The morphological changes of brain tissue were observed by HE staining. Results: In the aging group, the number of errors was increased in the ability test of jumping platform. In the water maze test, the escape latency was prolonged, the swimming time of the platform quadrant was shortened, the number of swimming through the platform was decreased, the number of brain cells was significantly decreased, the indexes of brain index, thymus index and spleen index Decreased; IL-2, IFN-γ levels and T lymphocyte proliferation decreased; compared with the aging group, loofah extract (375 mg / kg, 750mg / kg) can significantly antagonize these changes. Conclusion: Loofah extract can improve the learning and memory of aging mice, while increasing the body’s immune activity, to delay the role of aging process.
