目的:探讨转录因子激活增强子结合蛋白4(TFAP4)在胃癌中的表达及对预后的影响,并观察TFAP4在胃癌细胞迁移和侵袭过程中发挥的作用。方法:通过生物信息学分析,获取TFAP4在胃癌中的表达及在胃癌患者的预后中所起到的作用。采用Transwell小室法及划痕实验检测TFAP4对胃癌细胞迁移和侵袭能力的影响。应用蛋白质印迹法(Western blot)检测敲低TFAP4后上皮-间充质转化(EMT)相关通路蛋白的表达变化。对独立样本采用n t检验。n 结果:基因表达谱交互分析(GEPIA2)网站对胃癌组织的RNA测序(RNA-seq)数据进行分析,结果显示TFAP4在胃癌组织中高表达(n t=32.070,n P<0.01),差异有统计学意义。生存分析(KM-PLOTTER)网站对TFAP4进行生物信息学分析,TFAP4为胃癌预后的危险因素。划痕实验结果显示,敲低TFAP4的实验组在划痕48 h后相较于对照组迁移距离更短。Transwell实验结果显示,敲低TFAP4的实验组在24 h内穿膜细胞数量显著低于对照组[(83.000±3.786)个比(186.700±4.055)个,n t=18.690,n P<0.05],差异有统计学意义。Western blot结果显示,敲低TFAP4的表达后,上皮间充质转化(EMT)通路相关蛋白N-钙黏蛋白及基质金属蛋白酶(MMP)-2的表达水平随之降低。n 结论:TFAP4在胃癌中高表达,和不良预后呈正相关,具有促进胃癌细胞迁移和侵袭的作用。“,”Objective:To explore the expression of transcription factor activating enhancer-binding protein 4 (TFAP4) in gastric cancer and its effect on the prognosis, and observe the role of TFAP4 in the migration and invasion of gastric cancer cells.Methods:Through bioinformatics analysis, the expression of TFAP4 in gastric cancer and its role in the prognosis of gastric cancer patients were studied. Transwell chamber method and scratch test were used to detect the effect of TFAP4 on the migration and invasion of gastric cancer cells. Western blotting was used to detect the expression changes of epithelial-mesenchymal transition (EMT)-related pathway proteins after knocking down TFAP4.Results:The RNA-seq data of gastric cancer tissues were analyzed by the gene expression profiling interactive analysis 2(GEPIA2) website, and the results showed that TFAP4 was highly expressed in gastric cancer tissues (n t=32.070, n P<0.01). The kaplan-meier plotter (KM-PLOTTER) website was used to conduct bioinformatics analysis on TFAP4, a risk factor for the prognosis of gastric cancer. The scratch experiment showed that the experimental group knocking down TFAP4 had a shorter migration distance than the control group after 48 h of scratching. The Transwell experiment showed that the number of transmembrane cells in the experimental group with TFAP4 knockdown was significantly lower than that of the control group within 24 h [(83.000±3.786) cells vs. (186.700±4.055) cells,n t=18.690, n P<0.05]. Western blotting results showed that after knocking down the expression of TFAP4, the expression levels of EMT pathway related proteins N-cadherin and matrix metalloproteinase (MMP)-2 decreased.n Conclusion:TFAP4 is highly expressed in gastric cancer, positively correlated with poor prognosis, and can promote the migration and invasion of gastric cancer cells.
