目的观察血管紧张素转换酶(ACE)下调对胰腺癌细胞株SW1990运动侵袭能力的影响。方法设计并体外合成靶向ACE的小干扰RNA,将其转染到SW1990细胞中,蛋白质印迹法检测转染前后细胞中ACE蛋白的表达,采用Transwell小室检测转染前后细胞运动侵袭能力的改变。结果 ACE siRNA能有效降低SW1990细胞中ACE的表达。抑制SW1990细胞中ACE的表达后,SW1990细胞的运动能力明显下降,穿过人工基底膜的细胞数显著低于对照组(P<0.05)。结论核糖核酸干扰下调ACE表达能明显抑制胰腺癌细胞SW1990在体外的运动侵袭能力。
Objective To investigate the effect of angiotensin converting enzyme (ACE) down regulation on the invasiveness of pancreatic cancer cell line SW1990. Methods Small interfering RNA targeting ACE was designed and synthesized in vitro. The transfected SW1990 cells were transfected with SW1990 cells. The protein expression of ACE was detected by Western blotting before and after transfection. The changes of cell motility and invasion were detected by Transwell chamber. Results ACE siRNA can effectively reduce the expression of ACE in SW1990 cells. After inhibiting the expression of ACE in SW1990 cells, the motor ability of SW1990 cells was significantly decreased, and the number of cells passing through the artificial basement membrane was significantly lower than that of the control group (P <0.05). CONCLUSION: Ribonucleic acid (siRNA) knockdown ACE expression significantly inhibits the invasiveness of pancreatic cancer cell line SW1990 in vitro.
