目的 :观察诱导分化剂苯乙酸对胶质瘤细胞C6增殖的抑制作用 ,及对抑癌基因p16的蛋白水平表达变化的影响。方法 :[3H] TdR掺入法测细胞增殖率 ;免疫组化SP法检测p16基因蛋白水平的表达。结果 :应用苯乙酸后 ,细胞CPM值降低 ;p16基因胞浆阳性表达显著升高。结论 :苯乙酸对胶质瘤细胞存在剂量依赖性抑制 ,增强了抑癌基因p16表达活性 ,促使C6细胞向凋亡方向转化
OBJECTIVE: To observe the inhibitory effect of phenylacetate on proliferation of C6 glioma cells and its effect on the protein expression of tumor suppressor gene p16. Methods: [3H] TdR incorporation method was used to measure cell proliferation rate; immunohistochemical SP method was used to detect the expression of p16 gene protein. Results: After using phenylacetic acid, the cell CPM value decreased; the positive expression of p16 gene cytoplasm increased significantly. CONCLUSION: Phenylacetate inhibits the growth of glioma cells in a dose-dependent manner and enhances the expression of tumor suppressor gene p16, leading to the transformation of C6 cells toward apoptosis
