目的探讨地黄煎水、醇提物对小鼠脾细胞和卵巢颗粒细胞作用的影响。方法地黄煎水提物、醇提物、香菇多糖、等量培养液分别作用于小鼠脾细胞和卵巢颗粒细胞。酶联免疫吸附法(ELISA)检测细胞上清液白介素1(IL-1)、雌二醇(E2)浓度,荧光实时定量(RT-PCR)检测雌激素受体(ERα)、IL-1受体(IL-1R)的m RNA表达,流式细胞仪检测细胞表面ERα、IL-1R的分子水平。结果地黄煎水提组、醇提组均可提高IL-1、E2与ERα、IL-1R的表达水平(P<0.05),且地黄煎水提组E2表达水平高于醇提组(P<0.05)。地黄煎水提组可使IL-1R m RNA的表达水平降低(P<0.05)。结论地黄煎水、醇提物对小鼠脾细胞和卵巢颗粒细胞有较好的调节作用,水提组疗效略好于醇提组。
Objective To investigate the effects of decoction of JianDianJing and alcohol extract on the effects of spleen cells and ovarian granulosa cells in mice. Method Decoction Jian extract, alcohol extract, lentinan, equal culture medium were applied to mouse spleen cells and ovarian granulosa cells. The concentrations of IL-1 and E2 in the supernatant of the cells were detected by enzyme-linked immunosorbent assay (ELISA), and the levels of ERα and IL-1 were determined by real-time quantitative reverse transcription polymerase chain reaction (IL-1R) m RNA expression, flow cytometry cell surface ERα, IL-1R molecular level. Results Dihuang Jianshui group and alcohol group could increase the expression of IL-1, E2 and ERα, IL-1R (P <0.05), and the E2 expression in Decoction group was higher than that in alcohol group (P < 0.05). Decoction Jianshui group can reduce the level of IL-1R m RNA expression (P <0.05). Conclusion Decoction of Jian Huang, ethanol extract of mouse spleen cells and ovarian granulosa cells have a better regulatory effect, water extraction group was slightly better than alcohol group.
