目的:观察平阳降压方对自发性高血压大鼠(SHR)肝阳上亢证模型的血压及肾素-血管紧张素-醛固酮系统中肾素(Renin)、血管紧张素Ⅱ(AngⅡ)、醛固酮(ALD)的影响,探讨其降低血压的作用途径与机制。方法:将24只雄性SHR随机分为4组,每组6只,各组均予附子汤连续灌胃4周,第5周开始,平阳降压方组每日灌服浓度为2g/m L的平阳降压方煎剂8m L/kg体质量,卡托普利组每日灌服浓度为0.5mg/m L的卡托普利混悬液8m L/kg体质量,SHR模型组每日灌服蒸馏水8m L/kg体质量代替治疗,均连续4周;SHR对照组全程每日灌服蒸馏水8m L/kg体质量。取正常Wistar大鼠6只为正常对照组,自由进食、进水,不做任何处理。观察各组大鼠血压变化及血浆Renin、AngⅡ、ALD含量。结果:(1)急性降压实验中,平阳降压方组及卡托普利组SHR血压均较模型组降低(P<0.05),2组间24h总的降压效果无显著性差异(P>0.05),平阳降压方组24h内血压变化较卡托普利组平稳(P<0.05);慢性降压实验中平阳降压方组及卡托普利组SHR血压均较模型组明显降低(P<0.05)。(2)SHR对照组及SHR模型组中Renin、AngⅡ、ALD水平均高于正常对照组(P<0.05),2组间差异无统计学意义(P>0.05),平阳降压方组与卡托普利组血浆中Renin、AngⅡ、ALD水平均较模型组显著降低,差异有统计学意义(P<0.05),2组间比较,差异无统计学意义(P>0.05)。结论:(1)平阳降压方对SHR肝阳上亢证有明显的急慢性降压作用。(2)平阳降压方对SHR肝阳上亢证的降压作用可能与降低RAAS中Renin、AngⅡ、ALD水平,抑制RAAS活性有关。
Objective: To observe the effect of Pingyang Jiangya Decoction on the blood pressure and the changes of Renin, Ang Ⅱ and Ang Ⅱ in spontaneously hypertensive rats (SHR) Aldosterone (ALD), to explore its mechanism of action and mechanism of lowering blood pressure. Methods: Twenty-four male SHRs were randomly divided into 4 groups with 6 rats in each group. The rats in each group were treated with aconite decoction continuously for 4 weeks. From the fifth week, the daily feed concentration of Pingyang Jiangya Decoction group was 2g / m L Of Pingyang antihypertensive decoction 8m L / kg body weight, captopril group daily captopril concentration 0.5mg / m L captopril suspension 8m L / kg body weight, SHR model group daily Irrigation with distilled water 8m L / kg body weight instead of treatment, were continuous 4 weeks; SHR control group throughout the day with distilled water 8m L / kg body weight. Six normal Wistar rats were taken as normal control group, fed freely and infused water without any treatment. The changes of blood pressure and the contents of Renin, AngⅡ and ALD in each group were observed. Results: (1) In acute antihypertensive experiment, SHR blood pressure of Pingyang hypotensive group and captopril group were lower than that of model group (P <0.05), and there was no significant difference between the two groups > 0.05). The change of blood pressure in Pingyang antihypertensive group was more stable than that in captopril group within 24 hours (P <0.05). The SHR blood pressure of Pingyang antihypertensive group and captopril group was significantly lower than that of model group (P <0.05). (2) The levels of Renin, AngⅡ and ALD in SHR control group and SHR model group were higher than those in normal control group (P <0.05), there was no significant difference between the two groups (P> 0.05) The levels of Renin, AngⅡ and ALD in plasma in Toptop group were significantly lower than those in model group (P <0.05). There was no significant difference between the two groups (P> 0.05). Conclusion: (1) Pingyang antihypertensive regimen has obvious acute and chronic antihypertensive effects on SHR hyperactivity of liver. (2) The hypotensive effect of Pingyang Jiangya Recipe on SHR hyperactivity syndrome may be related to the decrease of Renin, AngⅡ, ALD levels and the inhibition of RAAS activity in RAAS.
