[目的]掌握刺糖多孢菌生长代谢特性,为提高多杀菌素产量及发酵稳定性提供依据。[方法]对刺糖多孢菌斜面及种子生长规律、培养工艺及移种标准进行研究。[结果]确定斜面移种标准为平板单菌落移种第1代斜面,菌龄7 d;液体种子最佳培养工艺为添加30粒玻璃珠,温度30℃,初始pH值8.5;液体种子移种标准为生物量大于1.4 g/L、氨基氮75~80 mg/L、还原糖低于7 g/L、pH值低于7.5、种龄30~36 h。[结论]采用标准种子移种,多杀菌素产量提高58.5%,不同批次间产量差异小于5%。
[Objective] The research aimed to master the growth and metabolism characteristics of spinosad, and provide the basis for improving spinosad production and fermentation stability. [Method] The slanting and seed growth rules, culture techniques and transplanting standards of S. spinosa were studied. [Result] The standard of slant transplanting was determined as the 1st generation slant transplanting slab single colony, and the bacteria age was 7 d. The best culture process of liquid seeds was adding 30 glass beads, the temperature was 30 ℃ and the initial pH value was 8.5. The standard is that the biomass is more than 1.4 g / L, amino nitrogen is 75-80 mg / L, the reducing sugar is less than 7 g / L, the pH value is less than 7.5 and the seedling age is 30-36 h. [Conclusion] With the standard seed transplanting, the yield of spinosad increased by 58.5% and the yield difference among different batches was less than 5%.
