目的 探讨脂质体—硫代磷酸化修饰的C erbB2 反义脱氧寡核苷酸 (C erbB2 antisensephosphorothioatedoligodeoxynucleotides ,C erbB2 S ODNs)对卵巢癌细胞系C erbB2 癌基因表达及细胞增殖的影响。方法 采用流式细胞仪技术 ,3 H TdR掺入方法了解脂质体—C erbB2 S ODNs对卵巢癌细胞系SKOV3 C erbB2 癌基因表达及细胞生长 ,细胞周期的作用。结果 脂质体—C erbB2 S ODNs能降低C erbB2 的表达 ,并抑制细胞生长达 30 % ;脂质体—C erbB2 S ODNs较单用C erbB2 S ODNs的作用效率增强约 4 0倍。结论 C erbB2 癌基因的反义调控可能在卵巢癌基因治疗中有一定作用 ;脂质体能够有效地增强C erbB2 S ODNs的作用效率。
Objective To investigate the effect of C erbB2 antisensephosphorothioated oligodeoxynucleotides (C erbB2 S ODNs) on oncogene expression and cell proliferation in human ovarian cancer cell line C erbB2. Methods The effect of liposome-erbB2 S ODNs on the oncogene expression, cell growth and cell cycle of ovarian cancer cell line SKOV3 C erbB2 was studied by flow cytometry and 3 H TdR incorporation. Results Liposome-C erbB2 S ODNs decreased the expression of C erbB2 and inhibited the cell growth by 30%. The effect of liposome-C erbB2 S ODNs was enhanced about 40% more than that of C erbB2 S ODNs alone. Conclusion Antisense C erbB2 oncogene may play a role in ovarian cancer gene therapy. Liposomes can effectively enhance the efficiency of C erbB2 S ODNs.
