以牛鼻软骨为原料酶法制备软骨粉,采用稀碱液浸提和乙醇沉淀的方法提取硫酸软骨素(Chondroitin Sulfate,ChS),通过单因素和正交试验对碱解工艺进行优化,确定的最佳条件是:料水比1:6,NaOH浓度5%,温度40℃,浸提时间6h,在此条件下,提取率达到12.58%,葡萄糖醛酸含量为32.44%,硫酸软骨素纯度达82%。紫外光谱分析:样品在196nm处有多糖的吸收峰,未见蛋白质(280nm)与核酸(260nm)的特征吸收峰;红外光谱分析发现为典型的硫酸软骨素A吸收峰。
Chondroitin sulfate (ChS) was prepared by enzymatic digestion of bovine nasal cartilage, Chondroitin Sulfate (ChS) was extracted by dilute lye extraction and ethanol precipitation. The optimum conditions were determined by single factor and orthogonal experiments The optimum conditions were as follows: the ratio of material to water was 1: 6, the concentration of NaOH was 5%, the temperature was 40 ℃ and the extraction time was 6h. The extraction rate was 12.58%, the content of glucuronic acid was 32.44% and the purity of chondroitin sulfate was 82 %. UV spectral analysis: The sample has a polysaccharide absorption peak at 196 nm, no characteristic absorption peak of the 280 nm and the 260 nm. The infrared absorption spectrum of the sample shows a typical absorption peak of chondroitin sulfate A.
