目的观察补脾益气方对支气管哮喘大鼠肺组织Toll样受体4(TLR4)的影响。方法将36只Wistar大鼠随机分为对照组、哮喘组、治疗组,每组12只。哮喘组使用卵蛋白致敏和激发制备支气管哮喘模型,对照组使用等量生理盐水代替卵蛋白,治疗组在第1次致敏时以补脾益气方10g/kg灌胃治疗,每天1次,共21天。观察各组大鼠嗜酸性粒细胞(EOS)等炎性细胞的计数,并采用RT-PCR检测各组大鼠肺组织TLR4的mRNA表达水平,采用免疫组化检测肺组织TLR4的蛋白表达水平。结果哮喘组大鼠肺组织TLR4的mRNA及蛋白的表达均显著高于对照组和治疗组(P<0.01);哮喘组大鼠血液中EOS计数及其他炎性细胞百分比均显著高于对照组和治疗组(P<0.01)。结论支气管哮喘大鼠肺组织TLR4mRNA和蛋白的表达显著增强,补脾益气方能有效抑制其表达,并同时降低血液中EOS等炎性细胞的数量。
Objective To observe the effect of “Bu Pi Yi Qi Fang” on Toll like receptor 4 (TLR4) in bronchial asthmatic rats. Methods Thirty - six Wistar rats were randomly divided into control group, asthma group and treatment group, with 12 rats in each group. In the asthma group, bronchial asthma model was made by sensitization and challenge with ovalbumin, the control group was treated with the same amount of saline instead of ovalbumin, and the treatment group was given gavage once a day for 10 g / A total of 21 days. The numbers of eosinophils (EOS) and other inflammatory cells in each group were observed. The mRNA expression of TLR4 in the lung tissue of each group was detected by RT-PCR. The protein expression of TLR4 in lung tissue was detected by immunohistochemistry. Results The mRNA and protein expression of TLR4 in asthmatic rats were significantly higher than those in control and treatment groups (P <0.01). The numbers of EOS and other inflammatory cells in asthmatic rats were significantly higher than those in control and Treatment group (P <0.01). Conclusion The expression of TLR4 mRNA and protein in the lung tissue of bronchial asthmatic rats was significantly increased. BuBu Yi Qi Fang could effectively inhibit the expression of TLR4 mRNA and decrease the number of inflammatory cells such as EOS in the blood.
