大量证据表明下颌下腺中存在近30种生物活性多肽,因此有人提出下颌下腺具有内分泌样功能,可将它归属于弥散神经内分泌系统。啮齿类动物下颌下腺分泌的多种生物活性多肽,主要定位于颗粒曲管细胞的分泌颗粒内,而人和其它哺乳动物的下颌一腺没有颗粒曲管细胞,多肽的产生部位尚未见系统报道;人胎的下颌下腺内是否含有生长抑素(SS)和神经生长因子(NGF)由未见报道。为了探讨下颔下腺复杂的内分泌功能,本实验收集胎龄为16~35周的胎儿20例,死后2小时内4%多聚甲醛灌注固定,取下颌下腺作40Urn厚冰冻切片。分别用SS、NGF抗体行漂浮ABC法免疫组化染色,棕色反应呈色。用正常羊血清或PBS作阴性对照。结果显示在入股下颔下腺内16周未见SS和NGF免疫反应阳性(Li)细胞;22周可见少数纹状管上皮细胞呈SS或NGF-Li阳性;30周时SS或NGF-Li纹状管上皮细胞增多,SS—Li主要分布于细胞的顶部胞质中,细胞基部胞质是阴性;而NGF—Li则分布于整个细胞质;细胞核处淡染。晚泡细胞均为阴性。人胎下颔下腺中纹状管上皮细胞中SS对该细胞中共存的其他生物活性多防的分泌活动可能有一定的调节作用;而发育期的NGF具有诱导神经纤维定向生长的作用,NGF的存在具有促进和维持交感神经和一些感觉神经元的发育和功能完整性
Numerous evidences show that there are nearly 30 bioactive peptides in the submandibular gland, so it has been suggested that the submandibular gland has endocrine-like functions that can be assigned to the diffuse neuroendocrine system. Many bioactive peptides secreted by the submandibular glands of rodents mainly locate in the secretory granules of the granular tubule cells while the human and other mammalian mandibular glands do not have granular astrocytes and the production sites of the polypeptides have not been systematically reported. Whether or not somatostatin (SS) and nerve growth factor (NGF) are present in the submandibular gland of a human fetus has not been reported. In order to investigate the complex endocrine function of the hypothalamic gland, 20 fetuses of gestational age ranging from 16 to 35 weeks were collected in this study. 4% paraformaldehyde was infused and fixed within 2 hours after death. The submandibular gland was removed for 40Urn thick frozen sections. Respectively with SS, NGF antibody floating line ABC immunohistochemical staining, brown reaction was colored. Use normal goat serum or PBS as a negative control. The results showed that no SS and NGF immunoreactive (Li) cells were found in the subgutal gland at 16 weeks after admission. A small number of striated tubular epithelial cells were positive for SS or NGF-Li at week 22; SS or NGF-Li at 30 weeks The proliferation of tubular epithelial cells, SS-Li mainly in the top of the cytoplasm of cells, cell-based cytoplasm is negative; and NGF-Li is distributed throughout the cytoplasm; Late bubble cells are negative.
