目的 :检测细胞核因子κB(NF -κB)、VEGF在大肠癌组织中的表达并探讨其与大肠癌肿瘤血管生成的关系。方法 :应用免疫组织化学方法检测NF -κBp6 5及VEGF在 5 2例大肠癌组织及 10例正常大肠组织中的表达情况 ,同时观察肿瘤内微血管密度 (MVD)。结果 :5 2例大肠癌中 34例NF -κBp6 5阳性 ,阳性产物主要定位于肿瘤细胞浆。NF -κBp6 5表达与患者的性别、年龄、大肠癌组织学分型、淋巴结转移、Duκes’分期无关。 10例正常大肠组织中NF -κBp6 5无 1例阳性表达。NF -κBp6 5表达与VEGF、MVD表达呈显著正相关。结论 :NF -κBp6 5是一个大肠癌相关的癌蛋白 ,NF -κBp6 5对VEGF可能有正向调节作用 ,并影响大肠癌血管生成
OBJECTIVE: To detect the expression of nuclear factor kappa B (NF-κB) and VEGF in colorectal carcinoma and to investigate its relationship with tumor angiogenesis in colorectal cancer. Methods: Immunohistochemistry was used to detect the expression of NF-κBp65 and VEGF in 52 cases of colorectal carcinoma and 10 cases of normal colorectal tissues. The intra-tumor microvessel density (MVD) was also observed. Results: Of the 52 cases of colorectal carcinoma, 34 cases were positive for NF-κBp65, and the positive products mainly located in the cytoplasm of the tumor. The expression of NF-κBp65 was not related to the patient’s gender, age, histological type, lymph node metastasis and Dukes’ staging. There was no positive expression of NF-κBp65 in 10 normal colorectal tissues. The expression of NF-κBp65 was positively correlated with the expression of VEGF and MVD. CONCLUSION: NF-κBp65 is a colorectal cancer-associated oncoprotein. NF-κBp65 may have a positive regulatory effect on VEGF and may affect the angiogenesis of colorectal cancer
