目的:优化细胞破壁法提取宝乐果中Ixisode的工艺条件,并考察Ixoside抗氧化活性以及细胞毒性。方法:用响应面法(RSM)优化超高压提取法提取Ixoside的条件,应用其对自由基清除活性以及对Hep G2细胞和A549细胞的细胞毒性来考察其抗氧化活性及细胞毒性。结果:基于等高线和方差分析推测得到宝乐果中Ixoside的最佳提取工艺为:提取溶剂为46%的甲醇水溶液,提取温度为31℃和提取压力为850帕并显示出较低的DPPH清除活性和较低的细胞毒性。结论:用响应面法(RSM)优化高压提取法能提高Ixoside的提取率。
OBJECTIVE: To optimize the technology of extracting cell wall Ixisode from pomegranate and investigate the anti-oxidative activity and cytotoxicity of Ixoside. Methods: The extraction conditions of Ixoside were optimized by response surface methodology (RSM). The antioxidant activity and cytotoxicity of Ixoside against free radical scavenging activity and cytotoxicity to Hep G2 and A549 cells were investigated. Results: Based on the contour analysis and variance analysis, the best extraction process of Ixoside in the pomegranate fruit was as follows: the extraction solvent was 46% methanol aqueous solution, the extraction temperature was 31 ℃ and the extraction pressure was 850 Pa and showed a lower DPPH Clearance activity and lower cytotoxicity. Conclusion: Optimization of high-pressure extraction using Response Surface Methodology (RSM) can improve the extraction rate of Ixoside.
