目的:研究CD-TK双自杀基因联合染料木黄酮对鼠前列腺癌的体内治疗作用。方法:取鼠前列腺癌细胞RM-1接种于鼠C57BL/6背部皮下建立移植瘤模型。将荷瘤鼠随机分为对照组、术黄酮组、TK/DC治疗组及TK/CD+染料术黄酮联合治疗组进行相应处理,实验结束后比较各组肿瘤体积、全部肿瘤标本行常规病理检验及bcl-2免疫组化检测。结果:对照组肿瘤体积为(1008.73±126.73)mm~3.染料木黄酮组肿瘤体积为0359.06±53.17)mm~3,双自杀基因组肿瘤体积为(222.60±26.79)mm~3.两者联合时肿瘤体积为(25.31±9.24)mm~3。双自杀基因组鼠肿瘤体积比染料木黄酮组小(P<0.01).两者联合时肿瘤体积最小(P=0.00。病理切片中显示对照组几乎未见细胞坏死.而联合用药组肿瘤大片坏死;免疫组化检测肿瘤标本中bcl-2含量显示对照组显强阳性.而联合治疗组仅见微弱阳性。结论:联用染料木黄酮能明显增强CD-TK双自杀基因系统对前列腺癌的治疗作用。
Objective: To study the in vivo therapeutic effect of CD-TK double suicide gene combined with genistein on murine prostate cancer. Methods: Rat prostate cancer cells RM-1 were inoculated subcutaneously on the back of C57BL / 6 mice to establish a transplanted tumor model. Tumor-bearing mice were randomly divided into control group, flavonoids group, TK / DC treatment group and TK / CD + flavonoids dye treatment group for the corresponding treatment, after the end of the experiment comparing the tumor volume of all groups, all the tumor specimens routine pathological examination and bcl-2 immunohistochemistry. Results: The tumor volume of the control group was (1008.73 ± 126.73) mm 3, the tumor volume of genistein group was 0359.06 ± 53.17 mm 3, and the size of double suicide genome tumor was 222.60 ± 26.79 mm 3. The tumor volume was (25.31 ± 9.24) mm ~ 3. The tumor size of mice in double suicide genotype group was smaller than that in genistein group (P <0.01), and the tumor volume was the smallest when combined (P = 0.00) .The pathological results showed that there was almost no cell necrosis in the control group, Immunohistochemical detection of bcl-2 in tumor samples showed that the control group was significantly positive, while the combined treatment group only showed weak positive.Conclusion: Combination of genistein can significantly enhance the therapeutic effect of CD-TK double suicide gene system on prostate cancer.
