利用大鼠心脏灌注等容收缩模型,观察槐定对顿抑心肌功能、代谢及超徽结构的影响。实验分心肌顿抑组(组1)、槐定保护组(组2)和正常对照组。结果:组1左室压力峰值(LVSP),左心室压力最大上升速率(+dp/dt-max)及其最大下降速率(-dp/dt- max)均比其它两组显著减低;其冠脉流出液乳酸脱氢酶(LDH)在灌注过程中进行性升高,左心室组织超氧化物歧化酶(SOD)活力降低,丙二醛(MDA)含量升高,心肌细胞内钙含量增加;心肌超微结构呈可逆性损害。组2中上述变化程度明显减轻。说明槐定能促进顿抑心肌功能恢复,减轻心肌细胞膜及其超微结构的损害,其机理可能与提高SOD活力,减轻脂质过氧化反应及心肌细胞钙超载有关。
The rat cardiac perfusion iso-contraction model was used to observe the effect of Valtadine on the function, metabolism and hyper-empirical structure of Stunned Myocardium. The experiment was divided into myocardial stunning group (group 1), protecting group (group 2) and normal control group. RESULTS: The peak left ventricular pressure (LVSP), left ventricular pressure maximal ascent rate (+dp/dt-max) and the maximum descending rate (-dp/dt-max) were significantly lower in group 1 than in the other two groups; coronary efflux Liquid lactate dehydrogenase (LDH) was progressively elevated during perfusion. The activity of superoxide dismutase (SOD) in the left ventricle decreased, and the content of malondialdehyde (MDA) increased, and the intracellular calcium content in the myocardium increased. Microstructure is reversible damage. The degree of change in Group 2 was significantly reduced. The results showed that budi could promote the recovery of myocardial function and reduce the damage of myocardial cell membrane and its ultrastructure. The mechanism may be related to the increase of SOD activity, decrease of lipid peroxidation and calcium overload of myocardial cells.
