目的观察葛根芩连汤对UC大鼠结肠组织中MTL、P选择素及结肠粘膜超微结构的影响,探讨葛根芩连汤对UC大鼠的作用机制。方法将120只大鼠随机分为8组,采用三硝基本磺酸(TNBS)乙/醇法建立UC模型,分别给予葛根芩连汤及部分拆方药组连续灌胃4 W后停药,采用放射免疫法检测大鼠结肠组织中MTL的含量,采用ELISA法测定结肠组织P选择素水平,透射电镜下观察结肠组织超微结构的改变。结果对大鼠结肠组织中MTL及P选择素的含量检测显示:模型组明显高于其它各组,全方组、去黄芩组、去甘草组明显低于模型组(P<0.01),其次为去葛根组、去黄连组、去葛根黄连组(P<0.05)。透射电镜下观察到葛根芩连汤及部分拆方药组对UC大鼠结肠粘膜具有修复作用。结论葛根芩连汤及部分拆方药组对UC大鼠结肠组织中MTL、VIP影响最明显的药组是葛根、黄连、炙甘草的组合。葛根、黄连、炙甘草的组合具有较好的胃肠动力恢复及炎症解除作用。
Objective To observe the effect of Gegen Qinlian Decoction on the expressions of MTL, P-selectin and colonic mucosa ultrastructure in the colon of UC rats and the mechanism of action of Gegen Qinlian Decoction on UC rats. Methods 120 rats were randomly divided into 8 groups. UC model was established by TNBS / B method. Gegen Qinlian Decoction and some prescriptions were given to rats for 4 weeks. The contents of MTL in the colon tissue of rats were detected by radioimmunoassay. The levels of P-selectin in colon tissues were determined by ELISA. The ultrastructural changes of colon tissues were observed under transmission electron microscope. Results The contents of MTL and P-selectin in colon tissue of rats in model group were significantly higher than those in other groups (P <0.01) To puerarin group, to berberine group, to puerarin group (P <0.05). Under the transmission electron microscope, Gegenqinlian Decoction and some prescriptions for the treatment of colonic mucosa in UC rats were found to be repaired. Conclusion Gegen Qinlian Decoction and some Cun Fang Prescriptions have the most significant effect on the MTL and VIP in the colon of UC rats, which is the combination of Pueraria, Coptis and Zhigancao. Pueraria, Coptis, Baked Licorice combination has good gastrointestinal motility recovery and relieve inflammation.
