目的:研究辛伐他汀对冠心病患者外周血T淋巴细胞PD-L1表达的影响。方法:抽取冠心病患者外周血,并分别予PBS(对照组)及110.32μg/L、1100.32μg/L浓度的辛伐他汀温育12h。采用逆转录聚合酶链反应、流式细胞技术分别检测人外周血T淋巴细胞PD-L1mRNA及蛋白表达水平。结果:随着给药浓度的增加,人外周血T淋巴细胞PD-L1表达逐步降低,与对照组比较差异有统计学意义(P<0.05)。结论:PD-L1参与了辛伐他汀的免疫调节作用。
Objective: To study the effect of simvastatin on the expression of PD-L1 in peripheral blood T lymphocytes of patients with coronary heart disease. Methods: Peripheral blood was collected from patients with coronary heart disease and incubated with simvastatin (110.32μg / L and 1100.32μg / L respectively) in PBS (control group) for 12h. Reverse transcriptase-polymerase chain reaction and flow cytometry were used to detect the expression of PD-L1mRNA and protein in human peripheral blood T lymphocytes. Results: With the increase of drug concentration, the expression of PD-L1 in T lymphocytes of human peripheral blood decreased gradually, which was significantly different from the control group (P <0.05). Conclusion: PD-L1 is involved in the immunoregulation of simvastatin.
