目的:研究曲尼司特对糖尿病肾病大鼠肾脏保护作用,并探讨其可能的机制。方法:雄性SD大鼠30只,随机分为正常对照组、糖尿病肾病模型组、曲尼司特治疗组。观察各组大鼠24 h尿蛋白定量、血糖、血尿素氮、血肌酐及肾脏病理的变化。应用RT-PCR方法和Western-blot方法测定肾组织硫氧还蛋白(Trx)mRNA和蛋白水平的表达。结果:曲尼司特能减少糖尿病肾病大鼠的尿蛋白,减轻肾小球纤维化,糖尿病模型组肾组织中Trx表达下降,曲尼司特治疗组大鼠肾组织中Trx的表达上升。结论:曲尼司特可能通过上调糖尿病肾病大鼠组织中Trx的表达,调节细胞外基质的形成,从而减轻肾脏病理损害,起到保护肾脏的作用。
Objective: To study the protective effects of tranilast on the kidneys of diabetic nephropathy rats and to explore its possible mechanism. Methods: Thirty male SD rats were randomly divided into normal control group, diabetic nephropathy model group and tranilast treatment group. The changes of 24 h urinary protein, blood glucose, blood urea nitrogen, serum creatinine and renal pathology were observed in each group. The mRNA and protein levels of thioredoxin (Trx) in kidney were measured by RT-PCR and Western-blot. Results: Tranilast reduced the urinary protein in diabetic nephropathy rats and decreased the glomerular fibrosis. The expression of Trx in renal tissue decreased in diabetic model group and increased in renal tissue in tranilast treatment group. CONCLUSIONS: Tranilast may play a protective role on kidney by up-regulating the expression of Trx in tissues of diabetic nephropathy and regulating the formation of extracellular matrix.
