目的:研究不同配伍的附子人参在缺糖缺氧复糖复氧大鼠心肌细胞的作用。方法:把原代培养6日的心肌细胞分成对照组、模型组以及不同配伍比例的附参组和空白血清组。观察各组对原代心肌细胞活力的影响以及对超氧化物歧化酶(SOD)、丙二醛(MDA)、一氧化氮(NO)及乳酸脱氢酶(LDH)的影响。结果:在72小时之内,与对照组相比,模型组以及空白血清组的细胞活力明显降低;模型组以及空白血清组的心肌细胞SOD的活性明显减小;MDA含量以及NO的分泌量和LDH释放率明显上升。与模型组比较,各配伍附参组的细胞凋亡几率明显更低,差异均有统计学意义(P<0.05)。结论:附子人参的配伍能够通过抑制缺糖缺氧复糖复氧导致大鼠心肌细胞的损伤,维持正常的心肌作用。
Objective: To study the effect of different compatibility of aconite ginseng on cardiomyocytes in hypoxia and hypoxia-reoxygenation rats. Methods: The primary cultured 6-day cardiomyocytes were divided into control group, model group and different proportion of the reference group and blank serum group. The effect of each group on the viability of primary cardiomyocytes and their effects on the activities of superoxide dismutase (SOD), malondialdehyde (MDA), nitric oxide (NO) and lactate dehydrogenase (LDH) were observed. Results: Compared with the control group, the cell viability of model group and blank serum group decreased significantly in 72 hours. The activity of SOD in model group and blank serum group was significantly decreased. The content of MDA and NO secretion LDH release rate increased significantly. Compared with the model group, the apoptosis rate of each compatibility group was significantly lower, the difference was statistically significant (P <0.05). Conclusion: The combination of aconite and ginseng can maintain the normal myocardial function by inhibiting the hypoxia-induced hypoxia-reoxygenation-induced cardiomyocyte injury in rats.
