目的 检测成釉细胞瘤中Cyclin D1和CDK4的表达,探讨其在成釉细胞瘤中的作用及意义。方法 选取52例成釉细胞瘤,其中良性35例,恶性17例,另取10例正常口腔黏膜作对照研究。应用免疫组化SP方法和原位杂交技术检测52例成釉细胞瘤和10例对照的正常口腔黏膜Cyclin D1蛋白及其mRNA和CDK4蛋白的表达。结果 良性和恶性成釉细胞中Cyclin D1蛋白及其mRNA的表达均显著高于对照的口腔黏膜(p<0/01)。CDK4的表达在良性、恶性和对照的口腔黏膜各组之间均具有显著性差异(p<0.01)。复发组CDK4的阳性细胞率(76.26±20.36)显著高于未复发组(65.45±17.41)(p<0.01)。在性别、年龄、发病部位、瘤体大小等方面Cyclin D1和CDK4表达均无显著性差异(p>0.05)。Cyclin D1蛋白及其mRNA的表达呈显著的正相关关系(p<0.01)。结论 Cyclin D1和CDK4参与成釉细胞瘤的发生和发展,CDK4可作为判断预后的参考指标。
Objective To detect the expression of Cyclin D1 and CDK4 in ameloblastoma and to explore the role and significance of Cyclin D1 and CDK4 in ameloblastoma. Methods 52 cases of ameloblastoma, including 35 cases of benign and malignant in 17 cases, another 10 cases of normal oral mucosa as a control study. The expressions of Cyclin D1 protein, mRNA and CDK4 protein in normal oral mucosa of 52 patients with ameloblastoma and 10 controls were detected by immunohistochemical SP method and in situ hybridization. Results The expressions of Cyclin D1 protein and mRNA in benign and malignant ameloblasts were significantly higher than those in the control oral mucosa (p <0/01). CDK4 expression was significantly different between the benign, malignant and control oral mucosa groups (p <0.01). The positive rate of CDK4 in recurrence group (76.26 ± 20.36) was significantly higher than that in non-recurrence group (65.45 ± 17.41) (p <0.01). There was no significant difference in the expression of Cyclin D1 and CDK4 in gender, age, disease location and tumor size (p> 0.05). Cyclin D1 protein and its mRNA expression was significantly positive correlation (p <0.01). Conclusions Cyclin D1 and CDK4 are involved in the occurrence and development of ameloblastoma. CDK4 may be used as a reference index for prognosis.
