本研究应用核酸探针杂交技术检测球囊血管成形术后不同时期血管平滑肌细胞血小板源生长因子(PDGF)基因表达情况。结果表明,球囊血管成形术后8~12小时,PDGF-A 链基因表达达高峰,48小时后明显降低,接近术前水平,PDGF-B 链基因呈持续表达,在术后略有增加,至术后两周仍能检出。本结果提示 PDGF 可能参与血管成形术后平滑肌细胞增殖的调节,在术后再狭窄形成中起重要作用。
In this study, nucleic acid probe hybridization was used to detect the gene expression of platelet derived growth factor (PDGF) in vascular smooth muscle cells at different stages after balloon angioplasty. The results showed that PDGF-A chain gene peaked at 8-12 hours after balloon angioplasty and decreased significantly after 48 hours. Concurrent to the preoperative level, PDGF-B chain gene was persistently expressed and slightly increased after surgery, Two weeks after surgery can still be detected. The results suggest that PDGF may be involved in the regulation of vascular smooth muscle cell proliferation after angioplasty and play an important role in the formation of postoperative restenosis.
