目的检测癌-睾丸抗原NY-SAR-35基因在视网膜母细胞瘤(RB)中的表达,探讨其作为RB特异性免疫治疗靶抗原的可能性。方法利用逆转录-聚合酶链反应方法,检测NY-SAR-35基因在15例RB组织、12例非肿瘤病变视网膜和22例其他正常眼部组织(眼睑皮肤12例、眼肌5例、结膜5例)中的表达,随机抽取RT-PCR阳性产物直接进行DNA测序,并将其表达结果与临床相关指标(性别、年龄、病理分型、肿瘤大小、临床分期)进行统计学分析,以探讨这一类肿瘤抗原的临床意义。结果在15例RB组织中,NY-SAR-35基因的表达率为60%(95%CI:35.2%~84.8%);在非肿瘤病变视网膜组织和其他正常组织中,无一例表达NY-SAR-35基因;其表达与临床相关指标(性别、年龄、病理分型、肿瘤大小、临床分期)无关(P>0.05)。结论 NY-SAR-35基因可特异性地表达于RB中,可能成为RB特异性免疫治疗的靶抗原,具有进一步研究开发的前景。
Objective To detect the expression of cancer-testis antigen NY-SAR-35 in retinoblastoma (RB) and to explore its potential as a target antigen for RB-specific immunotherapy. Methods NY-SAR-35 gene was detected by RT-PCR in 15 cases of RB tissue, 12 cases of non-tumor retina and 22 cases of other normal ocular tissues (eyelid skin in 12 cases, ocular muscle in 5 cases, conjunctiva 5 cases). The RT-PCR positive products were randomly selected for DNA sequencing, and the results were compared with the clinical indicators (gender, age, pathological type, tumor size and clinical stage) The clinical significance of this type of tumor antigen. Results The NY-SAR-35 gene expression rate was 60% (95% CI: 35.2% -84.8%) in 15 RB tissues. No expression of NY-SAR was found in non-neoplastic lesion retina and other normal tissues -35 gene. The expression was not related with clinical related indicators (sex, age, pathological type, tumor size, clinical stage) (P> 0.05). Conclusion The NY-SAR-35 gene can be specifically expressed in RB and may be the target antigen for RB-specific immunotherapy. It has the prospect of further research and development.
