本文研究了离体诱变对大麦游离小孢子培养的影响。结果表明,大麦花药预培养36后,经500radγ射线辐照,加快了小孢子的出愈速度,对小孢子分裂启动的比率影响不大,对胚状和愈伤组织的形成则有不利影响,其出愈率为25.3%,比对照37.1%低,但该处理产生的胚状体有一定的绿苗分化能力。在挑取花药前,穗子经500radγ射线辐照,不仅延缓了出愈时间,而且严重降低了孢子分裂启动的比率,从而使胚状体和愈伤组织的形成数大幅度下降,出愈率从对照37.1%下降到2.8%。
In this paper, the effects of in vitro mutagenesis on free microspore culture of barley were studied. The results showed that after barley anther culture 36, irradiation of 500radγ ray accelerated the speed of microspore emergence and had little effect on the initiation of microspore division, which had an adverse effect on the formation of embryoid and callus, The outcropping rate was 25.3%, which was lower than 37.1% of the control, but the embryoid body produced by this treatment had some ability of differentiation of green plants. Before the anther was picked, the ear radiated with 500 radγ-rays not only delayed the recovery time, but also severely reduced the rate of sporulation initiation, resulting in a sharp drop in the number of embryoid bodies and callus formation. Control dropped from 37.1% to 2.8%.
