目的:研究鳖甲煎丸含药大鼠血清对人肝癌HEPG2细胞p53和bcl-2表达的影响。方法:将实验大鼠随机分为4组,生理盐水灌胃组、鳖甲煎丸煎剂灌胃组、西药组、中西医联合用药组。无菌条件下从大鼠后腔静脉采血,分离血清。将同组大鼠血清混匀,冷藏备用。待人肝癌HEPG2细胞贴壁生长状态良好,加入含药大鼠血清及对照组大鼠血清,培养后取材制作标本。对标本进行含药血清作用的Western-blot蛋白表达分析即p53蛋白表达和Bcl-2蛋白的表达。结果:鳖甲煎丸煎剂灌胃组、西药组、中西医联合用药组p53蛋白表达增强,与灌服生理盐水组相比较有明显差异,差别有统计学意义(P<0.05)。鳖甲煎丸煎剂灌胃组、西药组、中西医联合用药组p53蛋白表达增强,三者无明显差异(P>0.05)。从数据上看,含药大鼠血清对p53蛋白表达增强的作用,中西药结合用药组最好,鳖甲煎丸灌胃组次之,最后是西药组。鳖甲煎丸煎剂灌胃组、西药组、中西医联合用药组Bcl-2蛋白表达减弱,与灌服生理盐水组相比较有明显差异,差别有统计学意义(P<0.05)。鳖甲煎丸煎剂灌胃组、西药组、中西医联合用药组Bcl-2蛋白表达减弱,三者无明显差异(P>0.05)。从数据上看,含药大鼠血清对Bcl-2蛋白表达减弱的作用,中西药结合用药组最好,鳖甲煎丸灌胃组次之,最后是西药组。结论:鳖甲煎丸含药大鼠血清对人肝癌HEPG2细胞p53蛋白表达有增强作用,对Bcl-2蛋白表达有减弱作用。
Objective: To study the effect of BJJ serum on p53 and bcl-2 expression in human hepatoma HEPG2 cells. Methods: The experimental rats were randomly divided into 4 groups: normal saline group, Biejiaotui pill decoction group, western medicine group and combination of Chinese and Western medicine group. Blood was collected from the posterior vena cava in aseptic conditions and the serum was separated. The same group of rats serum mixed, refrigerated standby. Hepatocellular carcinoma HEPG2 cells adherent growth well, the drug-containing rat serum and control group were added to the serum after the preparation of specimens drawn. The expression of p53 protein and Bcl-2 protein in Western-blot was analyzed by Western-blot. Results: The expression of p53 protein in BJJ decoction group was significantly higher than that in normal saline group (P <0.05). Biejiaotui pill decoction gavage group, Western medicine group, Integrative Medicine group p53 protein expression increased, there was no significant difference between the three (P> 0.05). Data from the point of view, drug-containing rat serum on the enhanced expression of p53 protein, the combination of Chinese and Western medicine was the best, Biejiaowan pill group followed by the stomach, and finally the western medicine group. Bcl-2 protein expression of BJJ decoction group was significantly lower than that of saline group (P <0.05). There was significant difference between BJ-2 group and western medicine group. The Bcl-2 protein expression in BJJ decoction group was significantly lower than that in western medicine group and western medicine group. There was no significant difference among the three groups (P> 0.05). Data from the point of view, drug-containing rat serum on the expression of Bcl-2 protein decreased the role of combination of Chinese medicine and Western medicine was the best, Biejiaowu Pills stomach group followed by the Western medicine group. Conclusion: BJJ serum can enhance the expression of p53 protein in human hepatocellular carcinoma HEPG2 cells and attenuate the expression of Bcl-2 protein.
