目的检测Dickkopf1(DKK1)在肝癌组织中的表达。方法采用四种不同方法检测DKK1在肝癌组织和癌旁肝组织的表达。结果半定量逆转录聚合酶链反应(RT-PCR)方法检测显示,DKK1在4/5肝癌中高表达;适时-PCR(Real-time PCR)方法检测显示,DKK1在8/14肝癌中高表达;Western blot方法检测显示,DKK1在2/4肝癌中高表达;免疫组化方法检测显示,DKK1在46/79肝癌中高表达,但与AFP表达水平、分化程度、TNM分期等临床病理特征无关。四种方法检测结果均显示DKK1在肝癌组织的表达显著高于癌旁肝组织。结论DKK1在肝癌组织表达高于癌旁肝组织,可能与肝癌发生、发展相关。
Objective To detect the expression of Dickkopf1 (DKK1) in hepatocellular carcinoma. Methods Four different methods were used to detect the expression of DKK1 in hepatocellular carcinoma and para-cancerous liver tissue. Results The results of semi-quantitative reverse transcriptase-polymerase chain reaction (RT-PCR) showed that DKK1 was overexpressed in 4/5 of the liver cancer cells. Real-time PCR showed that DKK1 was overexpressed in 8/14 hepatocellular carcinoma, The results of Western blot showed that DKK1 was overexpressed in 2/4 hepatocellular carcinoma. Immunohistochemical staining showed that DKK1 was highly expressed in 46/79 hepatocellular carcinoma, but not related to the clinicopathological features of AFP expression, differentiation and TNM staging. The results of four methods showed that the expression of DKK1 in hepatocellular carcinoma was significantly higher than that in adjacent liver tissue. Conclusions The expression of DKK1 in HCC tissues is higher than that in adjacent liver tissues, which may be related to the occurrence and development of HCC.
