本文建立了一种用高效液相色谱(HPLC)测定紫色疣石磺中胆甾醇含量的方法。采用COSMOSIL 5C18-MS-Ⅱ,(4.6mm×250mm,5μm)色谱柱,以甲醇为流动相,流速为1.0mL/min,检测波长204nm,柱温35℃,进样量20μL;检测得到胆甾醇含量在0.102~0.510mg/mL范围内与峰面积值的线性关系良好,相关系数R2为0.9998,回收率范围在96.08%~100.98%,RSD为1.72%;胆甾醇出峰时间为13min。本方法灵敏度和精密度高,分析时间短,可快速检测紫色疣石磺中的胆甾醇。紫色疣石磺样品中胆甾醇的平均质量浓度为503.7mg/kg。
In this paper, a high performance liquid chromatography (HPLC) was established for the determination of cholesterol in purple warts. The mobile phase consisted of COSMOSIL 5C18-MS-Ⅱ, (4.6 mm × 250 mm, 5 μm) with methanol as the mobile phase at a flow rate of 1.0 mL / min. The detection wavelength was 204 nm and the column temperature was 35 ° C. The injection volume was 20 μL. Cholesterol The linear range of the peak area was 0.102 ~ 0.510mg / mL. The correlation coefficient R2 was 0.9998. The recovery ranged from 96.08% to 100.98% with RSD of 1.72%. The peak time of cholesterol was 13min.
