目的:观察失答剌知丸对脑缺血再关注大鼠海马区Notch1和Jagged1含量的影响。方法:将102只SD大鼠随机分为17组,每组6只,分别为:空白组,假手术组,模型组,尼莫地平组,失荅剌知丸高剂量组、中剂量组和低剂量组,其中后5组分别依据再灌注时间的不同分为1 d、3 d、7 d组。以上各组除空白、假手术组外,均采用线栓法制备局灶性脑缺血再灌注模型(MCAO)。其中空白组、假手术组、模型组均用生理盐水灌胃;尼莫地平组给予浓度为1.08 g/L灌胃;失荅剌知丸组则以浓度分别为4.5 g/m L、3.0 g/m L和1.5 g/m L灌胃;以上各组每天同一时间灌胃,2次/d。各组分别在缺血再灌注1 d、3 d、7 d各时间点取材,检测Notch1和Jagged1水平。结果:(1)空白组与假手术组对比没有统计学意义(P>0.05);(2)模型组和各药物组相较于空白组、假手术组均有统计学意义(P<0.05);(3)各药物组相较于模型组均有统计学意义(P<0.05);(4)尼莫地平组与失荅剌知丸低剂量组对比有统计学意义(P<0.05),与失荅剌知丸中剂量组对比没有统计学意义(P>0.05),与失荅剌知丸高剂量组对比差异性显著(P<0.01);(5)失荅剌知丸低、中、高剂量组,随着1 d、3 d、7 d时间点的延长,Notch1和Jagged1表达量依次呈上升趋势,其中尤以7 d组表达量最高。结论:失答剌知丸治疗脑缺血可能与自身药效和促进海马区Notch1和Jagged1表达水平有关。
Objective: To observe the effect of Mudao Pill on the content of Notch1 and Jagged1 in hippocampus of rats following cerebral ischemia. Methods: 102 SD rats were randomly divided into 17 groups with 6 rats in each group. The rats in the blank group, sham operation group, model group, nimodipine group and Zhulizhazhimai high dose group, middle dose group and Low dose group, the latter 5 groups were divided into 1 d, 3 d, 7 d group according to the different reperfusion time. All the above groups were given focal cerebral ischemia-reperfusion model (MCAO) except the blank and sham group. The rats in the blank group, the sham operation group and the model group were intragastrically infused with normal saline. The nimodipine group was intragastrically administrated at a concentration of 1.08 g / L, and the Guizhi Guzhi pill group was infused at a concentration of 4.5 g / m L and 3.0 g / m L and 1.5 g / m L gavage; the above groups were fed daily at the same time, 2 times / d. The rats in each group were drawn at 1 d, 3 d and 7 d after ischemia reperfusion, and the levels of Notch1 and Jagged1 were detected. Results: (1) There was no significant difference between the blank group and the sham operation group (P> 0.05); (2) Compared with the blank group and the sham operation group, the model group and each drug group had statistical significance (P <0.05) (P <0.05); (3) There was significant difference between each drug group and model group (P <0.05); (4) There was significant difference between low dose Nimodipine group (P> 0.05), but there was no significant difference between the two groups (P <0.01); (5) , High dose group, the expression of Notch1 and Jagged1 gradually increased with the prolongation of 1 d, 3 d and 7 d, especially in the 7 d group. Conclusion: The treatment of cerebral ischemia with Lyingaizhi Wan may be related to its own efficacy and promoting the expression of Notch1 and Jagged1 in hippocampus.
