实验利用LC-MS/MS和火焰原子吸收光谱法方法测定大鼠口服甘草次酸后血浆中甘草次酸浓度和血清中6种金属元素钙(Ca),铜(Cu),铁(Fe),钾(K),镁(Mg),钠(Na)浓度,并分析甘草次酸对血清中6种元素的影响。检测结果表明大鼠口服甘草次酸后血清中Na,Cu元素浓度与甘草次酸血药浓度变化趋势相同。血药浓度在给药后2 h达峰值,血清中Na,Cu元素在给药后4 h浓度有显著增加(P<0.05)。随着给药剂量的增大,血清中Na,Cu元素浓度升高,当给药剂量为400,200 mg.kg-1时Na,Cu元素与对照组相比有显著性升高(P<0.05),对其他元素浓度则无显著性影响。本实验研究探讨了口服甘草次酸引起大鼠血清中6种金属元素的变化,为甘草次酸在临床中使用的副作用提供了实验依据。
The contents of glycyrrhetinic acid and the levels of six metals (Ca, Cu, Fe) in the plasma of rats after oral administration of glycyrrhetinic acid were determined by LC-MS / MS and flame atomic absorption spectrometry Potassium (K), magnesium (Mg) and sodium (Na) were determined. The effects of glycyrrhetinic acid on 6 kinds of elements in serum were analyzed. The results showed that the concentrations of Na and Cu in serum of rats after oral administration of glycyrrhetinic acid were the same as those of glycyrrhetinic acid. The plasma concentration peaked at 2 h after administration, and the contents of Na and Cu in serum increased significantly at 4 h after administration (P <0.05). The concentrations of Na and Cu in serum increased with the increase of the dose of Na and Cu, and the contents of Na and Cu increased significantly (P <0.05) compared with the control group at 400 and 200 mg.kg-1, , No significant effect on the concentration of other elements. The experimental study of oral glycyrrhetinic acid caused changes in the serum of 6 kinds of metal elements in rats for glycyrrhetinic acid used in clinical side effects provided an experimental basis.
