目的:探讨高迁移率族蛋白1(HMGB1)对多形核白细胞(PMN)黏附和游出肺毛细血管内皮细胞(PCEC)的影响机制.方法:分离培养大鼠PMN和PCEC,分别加入0,10,100,1000和10 000μg/L浓度的rHMGB1与PCEC单层培养.记录PMN黏附率.在Bovden小室迁移模型中加入上述浓度的rHMGB1,观察PMN穿过PCEC的迁移率.将PMN与100μg/L的rHMGB1孵育,流式细胞仪检测其表面CD11a/CD18和CD11b/CD18的表达率.结果:随着rHMGB1浓度的增加,PMN的粘附率(2.5%±0.5%,5.1%±0.9%,10.7%±1.7%,14.6%±2.6%,25.4%±4.3%)和穿过PCEC的游出率(0%,1.1%±013%,613%±1.2%,12.4%±2.7%,14.2%±3.1%)逐渐增加,但对向下室的迁移率无影响,州MGB1可明显增加PMN表面CD11a/CD18和CD11b/CD18表达的阳性率(均P<0.05)。结论:HMGB1通过上调CD11a/CD18和CD11b/CD18的表达增加PMN对PCEC的黏附率和游出率。
OBJECTIVE: To investigate the effect of high mobility group box 1 (HMGB1) on the adhesion of polymorphonuclear leukocytes (PMN) and the pulmonary capillary endothelial cells (PCEC) in rats.Methods: PMN and PCEC were isolated and cultured, 10, 100, 1000 and 10 000 μg / L rHMGB1 and PCEC monolayers were cultured.The PMN adhesion rate was recorded.The rHMGB1 concentration was added into the Bovden cell migration model to observe the PMN migration through the PCEC.The PMN and 100μg / L rHMGB1, and the expression of CD11a / CD18 and CD11b / CD18 were detected by flow cytometry.Results: With the increase of rHMGB1 concentration, the adherence rate of PMN (2.5% ± 0.5%, 5.1% ± 0.9%, 10.7% ± 0.7%, 14.6% ± 2.6%, 25.4% ± 4.3%) and the rate of exiting PCEC (0%, 1.1% ± 013%, 613% ± 1.2%, 12.4% ± 2.7%, 14.2% ± 3.1 %), But had no effect on the downward mobility. State MGB1 significantly increased the positive expression rates of CD11a / CD18 and CD11b / CD18 on the surface of PMN (all P <0.05). CONCLUSION: HMGB1 can increase the adhesion and the rate of PMEC to PCEC by up-regulating the expression of CD11a / CD18 and CD11b / CD18.
