【目的】探讨大蒜素对人膀胱癌细胞株BIU87细胞凋亡的影响。【方法】MTT法检测大蒜素对BIU87细胞生长的影响;倒置显微镜、TUNEL法观察细胞形态变化,流式细胞仪检测细胞凋亡率和细胞周期的变化;半定量RT-PCR法检测凋亡相关基因bcl-2,bax,caspase-3mRNA表达的变化,比色法测定caspase-3活性变化。【结果】大蒜素能明显抑制BIU87细胞的生长,与大蒜素的浓度和作用时间有关,5mg/L大蒜素作用5d可以杀死50%以上的细胞;可诱导BIU87细胞凋亡,细胞周期阻滞在S-和G2期;凋亡相关基因bcl-2表达减少,bax无明显变化,caspase-3表达及活性增高。【结论】诱导细胞凋亡是大蒜素发挥抗瘤效果的一种重要机制;凋亡发生可能与细胞周期阻滞、bcl-2和caspase-3基因表达及活性变化有关。
【Objective】 To investigate the effect of allicin on the apoptosis of human bladder cancer cell line BIU87. 【Methods】 The effect of allicin on the growth of BIU87 cells was detected by MTT assay. The morphological changes of cells were observed by inverted microscope and TUNEL. The apoptosis rate and cell cycle were detected by flow cytometry. The apoptosis of BIU87 cells was detected by semi-quantitative RT-PCR. The changes of mRNA expression of bcl-2, bax and caspase-3 were detected by colorimetric assay. 【Result】 Allicin significantly inhibited the growth of BIU87 cells, which was related to the concentration of allicin and the action time. 5 mg / L allicin could kill more than 50% of cells after 5 days, induce apoptosis and cell cycle arrest of BIU87 cells In S- and G2-phase, the apoptosis-related gene bcl-2 expression decreased, bax no significant change, caspase-3 expression and activity increased. 【Conclusion】 Induction of apoptosis is an important mechanism by which allicin can exert antitumor effect. The occurrence of apoptosis may be related to the cell cycle arrest, the expression of bcl-2 and caspase-3 genes and the changes of their activities.
