背景与目的已有的研究表明硒具有防癌作用和抑制乳腺癌、前列腺癌细胞株增殖的作用,但有关甲基硒酸(MSA)是否具有抗肺癌的作用目前尚不清楚。本研究的目的是探讨MSA对人高转移大细胞肺癌细胞株L9981是否具有生长抑制及诱导凋亡的作用,并对其分子机制作初步探讨。方法应用体外细胞培养技术,以MSA处理L9981细胞株,应用台盼蓝计数法、克隆形成实验和流式细胞术检测MSA处理L9981细胞株前后细胞株体外增殖、克隆形成和凋亡水平的变化。应用流式细胞术检测MSA处理L9981细胞株前后细胞周期、细胞周期相关蛋白和凋亡相关蛋白表达水平的变化。结果①MSA在大于0.5μmol/L的浓度时可显著抑制L9981细胞株的增殖(P<0.05),细胞被阻滞于G0/G1期。②MSA浓度在2.5μmol/L时可诱导L9981细胞株进入凋亡(P<0.05)。③MSA在5μmol/L时可明显抑制L9981细胞株的克隆形成能力(P<0.05)。④MSA能显著上调P53、P21、Fas、FasL和Bax蛋白表达水平。结论①MSA可显著抑制人高转移大细胞肺癌细胞株L9981的体外增殖和克隆形成能力,并诱导细胞凋亡;②MSA的抗肺癌作用可能与其调控肺癌细胞株L9981细胞周期和细胞凋亡相关基因表达有关。
BACKGROUND & OBJECTIVE Studies have shown that selenium has anti-cancer effects and inhibits the proliferation of breast cancer and prostate cancer cell lines. However, it is not clear whether methyl selenite (MSA) has an anti-lung cancer effect. The purpose of this study was to investigate whether MSA can inhibit the growth and induce apoptosis of human high metastatic large cell lung cancer cell line L9981 and to explore its molecular mechanism. Methods The cell line L9981 was treated with MSA in vitro. The cell proliferation, colony formation and apoptosis in L9981 cells were detected by trypan blue counting, clonogenic assay and flow cytometry. Flow cytometry was used to detect the changes of cell cycle, cell cycle related proteins and apoptosis related proteins before and after MSA treatment. Results (1) MSA significantly inhibited the proliferation of L9981 cells (P <0.05) at a concentration higher than 0.5 μmol / L, and the cells were arrested at G0 / G1 phase. ② The concentration of MSA at 2.5μmol / L induced apoptosis in L9981 cells (P <0.05). ③MSA at 5μmol / L significantly inhibited the colony-forming ability of L9981 cells (P <0.05). MSA could up-regulate the expression of P53, P21, Fas, FasL and Bax. Conclusion ① MSA can significantly inhibit the proliferation and colony-forming ability of human LHC cell line L9981 and induce apoptosis; ② The anti-lung cancer effect of MSA may be related to the regulation of the cell cycle and the expression of apoptosis related genes in L9981 lung cancer cell line .
