人体淋巴细胞微核试验的应用常受培养时细胞分裂增殖程度的限制。辨别不清细胞是否经过分裂是试验误差的重要原因。本文报道一种改良的差别染色程序(harlequin staining procedure),以便按细胞核的颜色很容易地鉴别出经培养而分裂的细胞。实验证明,微核只出现在分裂过的细胞。因此,若除去未分裂的细胞数,将明显地提高微核试验的准确性。方法:取健康成人静脉血,经~(60)Co源体外照射。
The application of human lymphocyte micronucleus test is often limited by the degree of cell division and proliferation during culture. It is an important reason for the experimental error to distinguish whether the cells are divided or not. Here we report an improved harlequin staining procedure that allows for the easy identification of cultured and dividing cells by the color of the nucleus. Experiments show that micronuclei only appear in the dividing cells. Therefore, if the number of non-dividing cells is removed, the accuracy of the micronucleus test will be significantly improved. Methods: Healthy adult venous blood was irradiated by ~ (60) Co source in vitro.
