比较了荧光显微镜观察法,流式细胞仪检测法及TdT末端标记法检测高能X线及加热诱导人鼻咽癌细胞系的凋亡指数。结果发现CNE-2细胞系受照2、4、8、12及20Gy后三种检测方法的凋亡指数荧光法分别为15%、20%、31%、38%及48%;流式法为16.5%、20.1%、33%、42.3%及47.5%;TdT法为24.6%、30.0%、40.6%、50.1%及58.7%。我们使用TdT法作为加热诱导凋亡的检测方法,发现于41℃培养0.5、1、1.5、2.5h后凋亡指数分别为16.7%、24.5%、30.7%及43%。结果提示TdT末端标记法是比较敏感的检测方法,CNE-2细胞系是对放、热疗均较敏感的实验模型
Fluorescence microscopy, flow cytometry and TdT-labeling were used to detect apoptosis index of high-energy X-ray and heat-induced human nasopharyngeal carcinoma cell lines.
