目的研究胰腺癌患者胰腺组织中肝细胞核因子1α(HNF1α)的表达变化及意义,并探讨其临床诊断价值。方法选取本院2011年10月-2014年12月27对胰腺导管细胞癌组织及其正常癌旁组织,采用实时荧光定量PCR法、链亲和素-生物素-过氧化物酶复合物(SABC)免疫组织化学染色及蛋白质免疫印迹(Western blot)法检测HNF1α的表达情况;应用受试者曲线(ROC)分析HNF1αmRNA对胰腺癌的诊断价值。结果与对照组相比,胰腺癌组织HNF1α蛋白和mRNA的表达均明显降低(P<0.01);HNF1αmRNA诊断胰腺癌的ROC曲线下面积(AUC)为0.89,敏感度和特异度分别为91.30%和75.00%。结论胰腺癌患者肿瘤组织中HNF1α表达的显著下调可能参与了肿瘤的发生、发展过程,其有可能成为胰腺癌诊断和治疗的新靶点。
Objective To investigate the expression and significance of hepatocyte nuclear factor-1α (HNF1α) in pancreatic tissues of patients with pancreatic cancer and to explore its clinical value. Methods From October 2011 to December 2014,27 pancreatic ductal cell carcinoma tissues and their adjacent normal tissues were selected and analyzed by real-time fluorescence quantitative PCR, streptavidin-biotin-peroxidase complex (SABC ) Immunohistochemical staining and Western blot were used to detect the expression of HNF1α. The ROC curve was used to analyze the diagnostic value of HNF1α mRNA in pancreatic cancer. Results Compared with the control group, the protein and mRNA expression of HNF1α in pancreatic cancer tissues were significantly decreased (P <0.01). The area under the curve of ROC curve (AUC) of HNF1α mRNA in pancreatic cancer was 0.89, the sensitivity and specificity were 91.30% 75.00%. Conclusion The significant down-regulation of HNF1α expression in pancreatic cancer may be involved in the development and progression of the tumor, which may become a new target for the diagnosis and treatment of pancreatic cancer.
