目的观察蒿鳖养阴软坚方抗CCl4复合因素致大鼠肝纤维化的作用。方法 Wistar大鼠随机分为对照组,模型组,蒿鳖养阴软坚方低、中、高剂量组,复方鳖甲软肝片及秋水仙碱阳性对照组。CCl4复合因素法建立肝纤维化模型,比色法测定血清丙二醛(MDA)、总蛋白(TP)、白蛋白(ALB)的量及肝组织羟脯氨酸(Hyp)水平,明胶酶谱法检测肝组织基质金属蛋白酶2(MMP-2)及基质金属蛋白酶9(MMP-9)的活性,免疫组化法检测肝组织α-平滑肌肌动蛋白(α-SMA)的表达。结果与模型组相比,随着剂量的增加,蒿鳖养阴软坚方组大鼠血清MDA水平逐渐下降,血清TP、ALB的量逐渐增加,肝组织Hyp的量和MMP-2活性逐渐下降,MMP-9活性变化不明显,肝组织α-SMA的表达下降。结论蒿鳖养阴软坚方能够对抗CCl4复合因素诱发的大鼠肝纤维化。
Objective To observe the effects of Artemisia turtle Yangyin Ruanjian Fang against CCl4 complex induced liver fibrosis in rats. Methods Wistar rats were randomly divided into control group, model group, Artemisia turtle Yangyin soft Jian low, medium and high dose group, Fufang Biejia Ruangan tablets and colchicine positive control group. CCl4 composite factor method was used to establish the model of hepatic fibrosis. The levels of serum malondialdehyde (MDA), total protein (TP), albumin (ALB) and hydroxyproline (Hyp) The activity of MMP-2 and MMP-9 in liver tissues was detected by immunohistochemistry. The expression of α-smooth muscle actin (α-SMA) in liver tissue was detected by immunohistochemistry. Results Compared with the model group, the serum MDA level of Artemisia turtleii yin yinjianfang group decreased gradually with the increase of dose, the levels of serum TP and ALB gradually increased, the amount of Hyp and the activity of MMP-2 in liver tissue gradually decreased, MMP-9 activity did not change significantly, liver α-SMA expression decreased. Conclusion Artemisia turtle Yangyin Ruanjian Fang CCl4 composite factor-induced liver fibrosis in rats.
