目的分析分离于江苏地域的3株不同宿主源弓形虫YZ-1(ChineseⅠ基因型)、YZ-2(ChineseⅠ基因型)和KS(基因型Ⅰ型)株对ICR小鼠的毒力,初步探讨弓形虫鼠毒力与基因型的相互关系。方法采用5μm滤膜及CF-11纤维素柱两步过滤法分离纯化弓形虫YZ-1、YZ-2、KS、RH株的速殖子,经计数后稀释至一定浓度,分别以104、103、102、101、100个速殖子的剂量经腹腔感染ICR小鼠,统计其死亡率与平均存活时间,判定3个分离株的鼠毒力型。结果纯化后虫体回收率为37.84%~39.18%,3个分离株感染小鼠引起的鼠死亡率与国际标准株RH株一致,均为100%;小鼠感染弓形虫后的存活时间随攻虫剂量的增加而缩短,KS分离株的鼠平均存活时间与YZ-1分离株相一致,与RH株也相近;YZ-2分离株感染小鼠的平均存活时间与KS、YZ-1和RH株差异有统计学意义(P<0.05)。结论弓形虫YZ-1、YZ-2和KS分离株与RH株的鼠毒力相同,属强毒株型;在ChineseⅠ型弓形虫分离株间,鼠毒力存在差异。
Objective To investigate the virulence of YZ-1 (Chinese Ⅰ genotype), YZ-2 (Chinese Ⅰ genotype) and KS (genotype Ⅰ) strains isolated from three different hosts in Jiangsu Province to ICR mice Correlation between Toxoplasma virulence and genotypes. Methods Tachyzoites of Toxoplasma gondii YZ-1, YZ-2, KS and RH strains were isolated and purified by two-step filtration using 5μm filter and CF-11 cellulose column. After counting, the tachyzoites were diluted to a certain concentration, , 102,101,100 tachyzoites were intraperitoneally infected with ICR mice, the mortality and the average survival time were counted to determine the virulence of the three isolates. Results The recoveries of parasites were 37.84% -39.18% after purification. The mortality of mice infected with the three isolates was 100% of that of the international standard strain RH strain. The survival time of mice infected with Toxoplasma gondii was The average survival time of KS isolates was consistent with that of YZ-1 isolate and RH strain. The mean survival time of mice infected with YZ-2 isolate was significantly lower than that of KS, YZ-1 and RH The difference was statistically significant (P <0.05). Conclusion Toxoplasma gondii YZ-1, YZ-2 and KS isolates have the same virulence as RH strains, and belong to virulent strains. There is a difference in virulence between Toxoplasma gondii isolates.
