目的探讨踝蛋白1(Talin-1)在不同前列腺癌细胞株中的表达情况和侵袭能力的关系。方法体外培养不同人前列腺癌细胞株(DU-145、PC-3、LNCaP)及人正常前列腺上皮细胞株(RWPE-1)。传代培养后利用MTT法检测各细胞株增殖能力。细胞划痕实验,Transwell小室侵袭和迁移实验检测各细胞株侵袭及迁移能力。RT-PCR技术检测各细胞株中Talin-1的mRNA表达水平。Western-blot技术检测各细胞株中Talin-1的蛋白表达水平。结果与RWPE-1细胞株相比,LNCaP细胞株、PC-3细胞株、DU145细胞株的增殖、侵袭和迁移能力明显增高(P<0.05),其中LNCaP、PC-3、DU-145细胞株的增殖、迁移以及侵袭能力依次递增(P<0.05)。前列腺癌LNCaP细胞株、PC-3细胞株、DU145细胞株中的Talin-1 mRNA及蛋白表达水平显著高于RWPE-1细胞株(P<0.05);其中LNCaP、PC-3、DU-145细胞株中的Talin-1 mRNA以及蛋白表达水平依次递增(P<0.05)。结论Talin-1与前列腺癌的增殖、侵袭和迁移能力相关;低表达Talin-1的前列腺癌的增殖、侵袭和迁移能力低,恶性度低;高表达Talin-1的前列腺癌的增殖、侵袭和迁移能力强,恶性度高。Talin-1可作为前列腺癌恶性度的预测指标之一。
Objective To investigate the relationship between the expression of Talin-1 and the invasiveness in different prostate cancer cell lines. Methods Different human prostate cancer cell lines (DU-145, PC-3, LNCaP) and human normal prostate epithelial cell line (RWPE-1) were cultured in vitro. After subculture, the proliferation of each cell line was detected by MTT assay. Cell scratch assay, Transwell chamber invasion and migration assay were used to detect the invasion and migration of each cell line. The mRNA expression of Talin-1 in each cell line was detected by RT-PCR. Western-blot was used to detect the protein expression of Talin-1 in each cell line. Results Compared with RWPE-1 cell line, the proliferation, invasion and migration of LNCaP, PC-3 and DU145 cell lines were significantly increased (P <0.05). LNCaP, PC-3 and DU-145 cell lines The proliferation, migration and invasive ability increased in turn (P <0.05). The expression of Talin-1 mRNA and protein in prostate cancer cell line LNCaP, PC-3 and DU145 was significantly higher than that in RWPE-1 cell line (P <0.05). LNCaP, PC-3 and DU-145 cells The expression of Talin-1 mRNA and protein in the strain increased in turn (P <0.05). Conclusions Talin-1 is associated with proliferation, invasion and migration of prostate cancer. Prostate cancer with low expression of Talin-1 has low proliferation, invasion and migration capacity and low malignant degree. Prostate cancer with high expression of Talin-1 is associated with proliferation and invasion and Migration ability, high degree of malignancy. Talin-1 can be used as a predictor of the malignancy of prostate cancer.
