目的:应用蛋白质组学技术分析筛选胆囊癌患者外周血清中生物标记蛋白,初步探讨其对胆囊癌早期诊断的意义。方法:应用表面加强激光解吸电离飞行时间质谱(SELDI-TOF-MS)技术,对45例胆囊癌患者及50例对照者(健康体检者)外周血清蛋白质进行检测,应用蛋白质芯片软件对蛋白质谱进行分析,建立诊断模型,并进行模型验证。结果:胆囊癌患者外周血清蛋白质表达谱与对照组相比,(5913Da,6181Da,13752Da)3个蛋白质荷比峰(M/Z)具有显著性差异,结果表明,该方法的灵敏度为86.7%,特异性为93.3%,这3个蛋白质峰数据建立蛋白质谱模型,用样本进行盲法测试。其敏感性为80%,特异性为90%。结论:应用SELDI-TOF-MS技术可有效筛选胆囊癌患者血清中特异性蛋白标记物,为胆囊癌患者的诊断提供了一条可靠的高灵敏度及特异性的方法。
OBJECTIVE: To analyze the biomarkers of peripheral blood serum of patients with gallbladder cancer by proteomics technology and to explore its significance in the early diagnosis of gallbladder cancer. Methods: Peripheral blood serum proteins of 45 patients with gallbladder carcinoma and 50 healthy controls were detected by SELDI-TOF-MS. Proteins were analyzed by protein chip software Analyze, establish diagnostic model, and model validation. Results: Compared with the control group, the protein expression profiles of peripheral blood serum in patients with gallbladder cancer (5913Da, 6181Da, 13752Da) were significantly different (M / Z). The results showed that the sensitivity of this method was 86.7% The specificity was 93.3%. The three protein peak data were used to establish the protein spectrum model, and the samples were tested blindly. Its sensitivity is 80%, specificity is 90%. Conclusion: The SELDI-TOF-MS technique can effectively screen the serum specific protein markers in patients with gallbladder cancer, providing a reliable and sensitive method for the diagnosis of gallbladder cancer patients.
