目的 :研究体外转染外源性野生型 p16基因对兔血管平滑肌细胞生长的影响。 方法 :构建野生型 p16基因的重组逆转录病毒载体 ,并体外转染兔血管平滑肌细胞 ,应用 Northern blot及 Westernblot检测外源 p16基因的表达 ,并用四唑盐 ( MTT)法及流式细胞仪检测其对细胞生长的抑制作用。 结果 :逆转录病毒载体可有效地将外源性 p16基因导入平滑肌细胞 ,表达 P16蛋白 ,并显著抑制血管平滑肌细胞的生长使细胞滞留于 G1 期。 结论 :体外转染外源性野生型 p16基因可有效抑制血管平滑肌细胞的生长。
Objective: To study the effect of transfection of exogenous wild-type p16 gene on the growth of rabbit vascular smooth muscle cells. Methods: Recombinant retroviral vector containing wild-type p16 gene was constructed and transfected into rabbit vascular smooth muscle cells in vitro. The expression of exogenous p16 gene was detected by Northern blot and Western blotting. The expression of p16 gene was detected by MTT assay and flow cytometry Its inhibitory effect on cell growth. Results: The retroviral vector could effectively induce the exogenous p16 gene into smooth muscle cells and express P16 protein, and significantly inhibit the growth of vascular smooth muscle cells so that the cells remain in the G1 phase. Conclusion: In vitro transfection of exogenous wild-type p16 gene can effectively inhibit the growth of vascular smooth muscle cells.
