采用四氮唑蓝(MTT)分析检测10例骨肉瘤细胞对氨甲喋呤(MTX)、阿霉素(ADR)、顺铂(CDDP)、长春新碱(VCR)、氮芥(NM)、丝裂霉素(MMC)的药物敏感性。其样本的检测率达9O%。骨肉瘤细胞对化疗药物的敏感性不一,其50%细胞生存的药物浓度(IC50)差异很大。在临床用药剂量下,6种化疗药物的平均生存率之间差异有显著性(P<0.01),其中ADR除与CDDP之间差异无显著性外,与其它相比差异有显著性。同时观察到6种药物的剂量效应曲线也不相同。因而认为此结果不仅可作为一种药物敏感性试验的方法指导临床选择化疗药物,而且提示骨肉瘤细胞的药物作用模式。
Methotrexate blue (MTT) assay was used to detect 10 cases of osteosarcoma cells for methotrexate (MTX), adriamycin (ADR), cisplatin (CDDP), vincristine (VCR), nitrogen mustard (NM), mitomycin The drug sensitivity of prime (MMC). The sample detection rate reached 90%. Osteosarcoma cells have different sensitivities to chemotherapeutic drugs, and their 50% cell survival drug concentration (IC50) varies greatly. The average survival rate of the six chemotherapeutic agents was significantly different (P<0.01) with the dose of clinical drugs. Therefore, it is considered that this result can not only be used as a method of drug sensitivity test to guide the clinical selection of chemotherapeutic drugs, but also suggests the mode of drug action of osteosarcoma cells.
