目的比较纯粹烤烟和白肋烟的致突变作用和细胞毒性,为开发低危害卷烟提供参考依据。方法以2种纯粹烤烟(F1,F2)和2种纯粹白肋烟(B1,B2)为研究对象,应用鼠伤寒沙门杆菌TA98和TA100菌株,分别在代谢活化和非活化条件下进行细菌回复突变试验;应用人淋巴母细胞TK6细胞进行TK基因突变试验,并通过测定相对悬浮生长率和平板效率考察细胞毒性,应用中国仓鼠成纤维细胞(CHL)细胞进行中性红试验。结果细菌回复突变试验结果表明,在非代谢活化条件下,4个样品均未检测出致突变作用;在代谢活化条件下,只有白肋烟样品被检测出具有较弱致突变作用。TK基因突变试验结果显示,与阴性对照组相比,白肋烟样品(B2)显著诱导突变频率的增加,差异具有统计学意义。TK6细胞细胞毒性试验和CHL细胞中性红试验结果表明,未见烤烟和白肋烟的细胞毒性之间差异有统计学意义。结论在该实验条件下,认为白肋烟的致突变作用强于烤烟。
Objective To compare the mutagenicity and cytotoxicity of pure flue-cured tobacco and burley tobacco, and to provide a reference for the development of low-risk cigarettes. Methods Tobacco two kinds of pure (F1, F2) and two pure Burley (B1, B2) for the study, application of Salmonella typhimurium strains TA98 and TA100, respectively and the bacteria in the metabolic activation of non-activation conditions revertant test; application TK6 human lymphoblastoid cells TK gene mutation test, and investigated by measuring the cytotoxicity relative survival rate and plating efficiency, application of Chinese hamster fibroblast (CHL) cells were neutral red test. Results Bacterial reverse mutation test results showed that, under non-metabolic activation condition, 4 samples were not detected mutagenesis; under metabolic activation conditions, the sample is detected only with burley weak mutagenicity. TK gene mutation test results showed that compared with the negative control group, Burley tobacco samples (B2) significantly increased the mutation frequency was significantly increased, the difference was statistically significant. TK6 cell cytotoxicity test and CHL cell neutral red test results showed that there was no difference between the cytotoxicity of flue-cured tobacco and burley tobacco was statistically significant. Conclusion Under the experimental conditions, burley tobacco is considered to be more mutagenic than burley tobacco.
